SIN Transit Question
While there are buses from Singapore to Melaka, you might be arriving a bit late. There is a listing bus services here (http://melaka.net/expressbus.htm) between Singapore and Melaka and it looks like only the Johara Express goes on late enough for your late arrival. I cannot find an on-line timetable. Suggest you phone.

times plus pick up locations included - last bus at 4pm, so you're out of luck with these guys (note this is at odds with the site KenJohn has linked to, but that des say last updated July 2003)
Just as a thought though, have you considered flying straight up to KL from Singapore then going to Melaka on the way back down?
The other option is to get a share taxi, either from Singapore (I think the malaysia ones have to go from a rank on Rochor Road) or again from JB. Any up to date backpacker type guidebook will have details here and you can hire the whole taxi no problem - rates are surprisingly cheap!

Normal Singaporean taxis are not allowed to cross into Malaysia (and vice versa). Specially licensed Singaporean taxis permitted to go to a single point in JB can be booked by phone (Johor Taxi Service from Singapore, tel. +656296-7054, SG$ 45.00 one way to fixed point in Johor Baru, Kotaraya), while Malaysian taxis (which can go anywhere in Malaysia) can be taken from Rochor Rd at premium rates. A combination ride from anywhere in Singapore to anywhere in Malaysia can also be arranged, but you'll need to swap cabs halfway through: this will cost S$40 and up, paid to the Singaporean driver. In the reverse direction towards Singapore, you can take taxis from Kotaraya to any point in central Singapore (S$30) or Changi Airport (S$40).
You can also opt to share taxis from Rochor Rd, which usually works out to around S$7 per person with four people. The main advantage here is that you don't need to lug your stuff (or yourself) through Customs at both ends; you can just sit in the car.
The most expensive option is to take a limousine taxi specially licensed to take passengers from any point to any destination, but only a few are available and they charge a steep RM150 per trip. Advance booking is highly recommended, tel. +607-5991622.

If you're determined to get to Melaka that night, your best bet might be to call Johor taxi service (number above) and ask them to pick you up at SIN (note, i'm not 100% sure if they can do this) then get a share taxi over to Melaka once in JB.
Cost for that appears as if it is going to be in the region of £25 so that'll be US$45 or so (for the whole taxi!) but it's going to be very late by the time you get to Melaka...
If you want to minimise cost and avod a night in Singapore, you could always get to JB then check into a hotel there - the 5* places are very reasonably priced - then continue by taxi or bus the next day.
